Mitochondrial-derived peptides

Small open reading frames within mitochondrial DNA encode short peptides — humanin, MOTS-c and the SHLP family among them — that appear to participate in retrograde signalling to the nucleus. MOTS-c has been studied in relation to AMPK activation, glucose handling and exercise adaptation, largely in cell and rodent systems with a small number of human observational measurements.

Mitochondria-targeting synthetic peptides

SS-31, also known as elamipretide, binds cardiolipin in the inner mitochondrial membrane. Cardiolipin organises the respiratory supercomplexes, and its remodelling is a recognised feature of mitochondrial dysfunction. This compound has progressed into registered human clinical trials in primary mitochondrial disease, which places it further along the evidence hierarchy than most peptides on this site — with mixed trial results that should be read directly rather than summarised second-hand.

Why mitochondrial endpoints are hard to measure

Meaningful mitochondrial research requires functional measures — respirometry on isolated mitochondria or permeabilised fibres, ATP production rates, membrane potential, reactive oxygen species production. Studies reporting only mitochondrial content markers, or subjective energy outcomes, do not demonstrate improved mitochondrial function.
