Multi-receptor metabolic pharmacology

The move from one receptor to two to three has been the dominant story of the last five years. Attention is now turning to what combined receptor engagement does to composition of weight change, to energy expenditure specifically, and to tolerability — questions single-endpoint trials were not designed to answer.

Delivery beyond injection

Oral peptide formulation using absorption enhancers has been demonstrated to be feasible, though bioavailability remains low and variable. Intranasal, transdermal and implantable approaches are all under active study, and delivery is increasingly the limiting factor rather than the pharmacology.

Computational design

Structure-prediction and generative design tools have reduced the cost of proposing novel sequences dramatically. The bottleneck has shifted downstream to synthesis, characterisation and validation, which are unchanged in cost — so the volume of proposed sequences now far exceeds the volume that will ever be tested.
