Ipamorelin and CJC-1295: Growth Hormone Secretagogue Research

Growth hormone (GH) secretion is governed by the hypothalamic-pituitary axis through the opposing actions of growth-hormone-releasing hormone (GHRH) and somatostatin, modulated further by the ghrelin/GHS-R1a pathway. Research peptides in this space allow investigators to probe each arm of this regulatory system independently.

Ipamorelin is a selective pentapeptide ghrelin-receptor agonist developed to stimulate GH release without the cortisol or prolactin spikes seen with earlier GHRPs such as GHRP-6. This selectivity makes it a useful research tool for isolating GHS-R1a signalling without confounding HPA-axis effects.

CJC-1295 is a tetrasubstituted GHRH(1-29) analogue. The variant with a DAC (Drug Affinity Complex) moiety binds albumin and extends half-life to multiple days, while the no-DAC variant retains a half-life closer to native GHRH. Pairing CJC-1295 with Ipamorelin in research models produces a synergistic GH pulse — the GHRH analogue raises baseline pituitary readiness while the ghrelin mimetic triggers release.

Published rodent and in-vitro work examines pulsatility, IGF-1 downstream signalling and tissue-specific GH receptor expression. Because the GH pulse is sensitive to feeding state, sleep cycle and stress, well-controlled protocols and accurate dosing are essential. Lyophilised reference standards with HPLC verification reduce one source of experimental variability.

Researchers commonly include Sermorelin (a shorter GHRH(1-29) analogue) and Tesamorelin as comparison standards in this class. MK-677 (Ibutamoren), although a small-molecule rather than a peptide, is also referenced in the secretagogue literature as an orally bioavailable GHS-R1a agonist.
